Genmab and Incyte have announced a clinical trial collaboration to evaluate a new combination treatment for patients with first-line KRAS G12D-mutated metastatic colorectal cancer (mCRC). The planned Phase 1/2 study will combine Genmab’s investigational bispecific antibody petosemtamab, which targets EGFR and LGR5, with Incyte’s investigational KRAS G12D inhibitor INCB161734 and standard-of-care chemotherapy.
The combination brings together two complementary approaches: targeting EGFR and LGR5 pathways involved in tumor growth and cancer stem-like cells, while directly inhibiting the KRAS G12D mutation. KRAS mutations occur in approximately 40% of colorectal cancers, with G12D being the most common KRAS variant, making it an important target for new treatment strategies.
Genmab will conduct the study, while Incyte will co-fund the trial and supply INCB161734. Both companies aim to investigate whether the combination can offer a new first-line treatment option for this difficult-to-treat patient population.
